"Best" and "favourite" are not the same thing.

Best would probably be Watchmen, Sandman, or Maus. I don't particularly enjoy any of those.

Favourite, for me, is going to be Excalibur. The original Claremont run, and then the far more recent Captain Britain and MI:13 series (which is just Excalibur by another name) are some of the most fun superhero books ever created.

If I had to choose ONE comic book, maybe FF Annual #3 for the wedding story packed with Lee/Kirby fun, even though I think FF #51 is a better story, but my favorite story arc is Avengers #164-#166, and favorite run is Avengers #164-#176.

The Death of Captain Marvel, is one of my all time favorite stories. It is moving because we are familiar with the way cancer destroys lives. This was a very touching and unforgettable story for me.
